Expression of DOG-1 protein is elevated in the gastrointestinal stromal tumors (GIST s), c-kit signaling-driven mesenchymal tumors of the GI tract. DOG-1 is rarely expressed in other soft tissue tumors, which, due to appearance, may be difficult to diagnose. Immunoreactivity for DOG-1 has been reported in 97.8 percent of scorable GIST s, including all c-kit negative GIST s. Overexpression of DOG-1 has been suggested to aid in the identification of GISTs, including Platelet-Derived Growth Factor Receptor Alpha mutants that fail to express c-kit antigen. Recognizes a protein of 83 kDa, which is identified as Factor XIIIa. It has been identified in platelets, megakaryocytes, and fibroblast-like mesenchymal or histiocytic cells in the placenta, uterus, and prostate, monocytes and macrophages and dermal dendritic cells. Anti-factor XIIIa has been found to be useful in differentiating between dermatofibroma (almost all cases are positive), dermatofibrosarcoma protuberans (-/ ) and desmoplastic malignant melanoma (-). Anti-factor XIIIa positivity is also seen in capillary hemagioblastoma, hemangioendothelioma, hemangiopericytoma, xanthogranuloma, xanthoma, hepatocellular carcinoma, glomus tumor, and meningioma. This antibody reacts with Ig domain of CD47 protein. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction. Caldesmon HMW is the high molecular weight variant of Caldesmon. Two closely related variants of human caldesmon have been identified which are different in their electrophoretic mobility and cellular distribution. The h-caldesmon variant (120'150 kDa) is predominantly expressed in smooth muscle whereas l-caldesmon (70'80 kDa) is found in non- muscle tissue and cells. Neither of the two variants has been detected in skeletal muscle. This antibody recognizes only the 150 kDa variant (h-caldesmon) in Western blots of human aortic media extracts and is unreactive with fibroblast extracts from cultivated human foreskin. Caldesmon is a developmentally regulated protein involved in smooth muscle and non-muscle contraction. The nm23 gene, a potential suppressor of metastasis, was originally identified by differential hybridization between two murine melanoma sub-lines, one with a high and the second with a low metastatic capacity.
